چکیده

Abstract Introduction Racemic ketoconazole (RK) is a steroidogenesis inhibitor used for treatment of Cushing’s syndrome. Levoketoconazole (COR-003), the pure 2S,4R enantiomer, potentially more potent and safe compared to RK. We in vitro effects levoketoconazole RK on adrenocortical pituitary adenoma cells. Materials methods HAC15 cells 15 primary human neoplastic cultures (+/− ACTH), murine (AtT20) corticotroph were incubated with or (0.01-10 µM). Cortisol ACTH measured using chemiluminescence immunoassay system, steroid profiles by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Results In HAC15, inhibited cortisol at lower concentrations (IC50: 0.300 µM) (0.611 µM; P < 0.0001). IC50 values basal production varied over 24-fold range (0.00578-0.140 µM), 2 patients having higher sensitivity vs (2.1- 3.7-fold). LC-MS/MS analysis selected cases revealed inhibition other profile components AtT20, cell growth secretion (10 µM: −54% −38%, respectively), number 1 (−44%, 0.001). Conclusion potently inhibits cells, variable degree suppression between specimens. adrenal might also inhibit Together previously reported potential advantages, this indicates that promising novel pharmacotherapy

investigating the feasibility of a proposed model for geometric design of deployable arch structures

deployable scissor type structures are composed of the so-called scissor-like elements (sles), which are connected to each other at an intermediate point through a pivotal connection and allow them to be folded into a compact bundle for storage or transport. several sles are connected to each other in order to form units with regular polygonal plan views. the sides and radii of the polygons are...

Medical Treatment of Cushings Syndrome: Adrenal-Blocking Drugs and Ketaconazole

Cushing’s syndrome is associated with serious morbidity and increased mortality. Irrespective of its cause, i.e. a pituitary adenoma, ectopic ACTH production or an adrenal neoplasia, Cushing’s syndrome is primarily treated surgically. However, when surgery is unsuccessful or contraindicated, medical therapy is needed to treat hypercortisolism. The spectrum of available drugs includes adrenal-bl...

Neuropsychiatric Disorders in Cushings Syndrome

Glucocorticoids are crucial in the initiation and consolidation of the stress response. Patients with active Cushing’s syndrome (CS) are exposed to excessive endogenous glucocorticoid levels. In these patients, psychopathology is often being observed. The most common co-morbid disorder is major depression, but to a lesser extent mania and anxiety disorders have also been reported. A severe clin...

Medical Treatment in Cushings Syndrome: Dopamine Agonists and Cabergoline

Dopamine (DA) is a catecholamine with a wide range of functions and whose five subtype receptors are found in different organs where they exert a mainly inhibitory action. Since this action may also appear in a number of secretory tumors in various locations, DA agonists have elicited some interest as a medical treatment for hypercorticism. Non-iatrogenic Cushing’s syndromes are due in 70% of t...
